SA Rugby has confirmed that de Bruin asked to be released from his role due to personal and medical reasons.
Coach Rassie Erasmus says he'll consider a number of options before naming a specialist replacement coach on a short-term basis.
Meanwhile, Springboks skipper Siya Kolisi will return from a knee injury against Argentina this weekend in Pretoria.
Kolisi hasn't played a test yet this year owing to the injury and is expected to fall well short of playing 80 minutes so won't be the on-field captain.
Those duties have been given to 38-year-old hooker Schalk Brits, who becomes the oldest first-time captain of the 'Boks.
